PY - 2020 DA - 2020/12/18 TI - Building a transgenic sexingstrain for genetic control of the Australian sheep blow fly Lucilia cuprina using two lethaleffectors JO - BMC Genetics SP - 141 VL - 21 IS - 2 AB - The sterile insect technique (SIT) has beensuccessfully used in many pest management programs worldwide. Some SIT programs release both sexes due to the lack of geneticsexing strains or efficient sex separation methods but sterilefemales are ineffective control agents. Transgenic sexingstrains (TSS) using the tetracycline-off control system havebeen developed in a variety of insect pests, from which femalesdie by either of two commonly used lethal effectors:overexpression of the transcription factor tetracycline transactivator (tTA)or ectopic expression of a proapoptotic gene, such as head involution defective(hid). The lethality fromtTA overexpression is thought to be due to “transcriptionalsquelching”, while hid causeslethality by induction of apoptosis. This study aims to createand characterize a TSS of Luciliacuprina, which is a major pest of sheep, bycombining both lethal effectors in a single transgenicstrain.
