Amplification of plasmid flanking sequences using semi-random two-step PCR (ST-PCR). In the first round of amplification (using primers 1.1 and 1.2), an 'adaptor' sequence is incorporated. In the second round (using primers 2.1 and 2.2), the flanking sequences are amplified.
